ABSTRACT
Pashmina (cashmere), fine fiber obtained from the Pashmina goat (Capra hircus) as its down fiber
(undercoat), is produced from the specialized structure of secondary hair follicle (SHF). Various regulatory genes have been alluded in induction and progression of the fiber developmental phases viz anagen (growth), catagen (involution), and telogen (resting). The present study was undertaken for structural and functional analysis of bone morphogenetic protein 2 (BMP2) in Pashmina goats. Comparative structure modeling was used to predict 3D structure of capra hircus BMP2 protein and various Bioinformatic tools were used for model evaluation and estimation of quality of predicted structure. GO annotation was used to capture the diverse biological functions attributed to BMP2 protein. Furthermore, we have studied expression of BMP2 gene at Anagen and Telogen stages of Pashmina fiber development in Pashmina (Changthangi) goats by performing quantitative Real Time PCR. Based on structural analysis, we obtained
C-score of -1.7, TM score of 0.51±0.15 and estimated RMSD value of 11.1±4.6Å for our predicted model. PROCHECK analysis showed 89.1% residues of BMP2 protein in favored and allowed region of Ramachandran plot. Moreover qRT-PCR data revealed a significant difference in expression pattern of BMP2 gene (higher at anagen stage) for Pashmina (Changthangi) goats of Ladakh region only. air follicle growth and coat development.
