Inter and Intra Chromosomal Karyotype Asymmetry Assessment in Barley (Hordeum vulgare L.)

ABSTRACT

Seeds of Hordeum vulgare cv. Azad and Jyoti obtained from Institute of Agricultural Sciences (IAS), B. H. U., Varanasi, Uttar Pradesh (India) were germinated. Somatic root tip cells were processed through conventional 2% acetocarmine staining squash technique for metaphase karyotype and photomicrographs analysis. Somatic chromosome number was counted 2n = 2x = 14 and karyotype asymmetry were classified as 1A and 2A for cv. Azad and Jyoti, respectively. Inter (chromatin length and arm ratio) and intra (AsK%, TF% and S%) chromosomal karyotype asymmetry indices suggested that cv. Azad was moderately symmetric than cv. Jyoti. Karyotype formula [2n = 14m (2sat)] was more symmetric for cv. Azad than cv. Jyoti [2n=12m (2 sat)+2sm]. Similarly, karyotype classification (1A) showed chromosome resemblance in cv. Azad than cv. Jyoti (2A).
